Ghrelin is a peptide hormone structure (28 amino acids) produced by the endocrine cells of the gastrointestinal mucosa (primarily P/D1 cells in stomach) as well as the epsilon-cells in the pancreas responsible for the stimulation of hunger.
It was isolated in 1999 from the stomach of the rat and described cicatricure as an endogenous ligand for the growth hormone secretagogue receptor (GHS-R) in the pituitary gland. Ghrelin stimulates growth hormone release in a different way GHRH (growth-hormone-releasing hormone).
From the chemical point of view ghrelin due to the unique structure of acylation of the serine cicatricure residue at position 3 of chain amino octanoic acid - Fatty acid - catalyzed process GOAT enzyme (ghrelin O-acyl transferase) recently identified. Octanoilarea is essential for the biological activity of the hormone.
Ghrelin is secreted in the form of a precursor - preprogrelina; posttranslational cicatricure modifications are generated from both acylated form - acyl-ghrelin - and non-acylated form - des-acyl ghrelin. In circulation are present both ghrelin, des-acyl, however predominates ghrelin, a form devoid of endocrine activity but has some cellular effects, according to recent studies.
Ghrelin receptor (a G-protein coupled receptor: GHS-R), cicatricure found ahead of its natural cicatricure agonist (1996), is known as the growth hormone secretagogue receptor. Acylated ghrelin binds to this receptor variant GHSR1a while des-acyl cicatricure ghrelin does not have this ability. GHSR1a is found in abundance in the hypothalamus and pituitary gland, but is also present in other areas of the central cicatricure nervous system cicatricure and in peripheral tissues, indicating that ghrelin exert other effects in addition to growth hormone secretagogue cicatricure activity. Thus, studies have shown that ghrelin stimulates appetite (central orexigenic effect) and food consumption occurs in the regulation of carbohydrate metabolism and energy homeostasis, stimulates gastric acid secretion and motility, reduces blood pressure and heart inotropismul improves endothelial function, stimulates the proliferation normal cicatricure cell lines (including cells belonging to the immune system), inhibits the secretion of pro-inflammatory cytokines and stimulate bone osteoblast proliferation and activity. Because cicatricure of these multiple effects, changes in the levels cicatricure of circulating ghrelin receptor expression and of GHSR1a has been implicated in the pathogenesis of several cicatricure diseases, such as growth hormone deficiency, wasting, anorexia, obesity, diabetes, metabolic syndrome, polycystic ovary syndrome, gastroparesis , gastric ulcer, atherosclerosis, hypertension, chronic heart failure, dilated cardiomyopathy, sepsis, osteoporosis. In these conditions cicatricure it is beneficial modulation of ghrelin, the hormone that could become a new therapeutic target and monitor its current cicatricure levels could have clinical utility 1, 4, 5.
Orexigenic cicatricure effect of ghrelin is due to actions exerted at hypothalamic arcuate and paraventricular nuclei level known to be involved in regulating appetite. Thus, ghrelin activates neurons containing orexigenic peptides cicatricure (NPY - neuropeptide Y and AgRP - agouti-related peptide) and to those containing peptides inhibit appetite suppressant (CART - cocaine and amphetamine-related transcript cicatricure and POMC - pro-opiomelanocortin). Leptin exerts opposite cicatricure effects on the same neurons, suggesting that these two hormones are complementary components of a regulatory system that informs the central nervous system connected with imbalances in the power acute and chronic. In turn, the neurons in the arcuate nucleus neurons send impulses to the lateral hypothalamic area containing the orexin, cicatricure thus stimulating appetite.
Alongside its role in initiating food intake, cicatricure ghrelin cicatricure occurs in adipose tissue accumulation (through direct promotion adipogenezei, creşeterea preference for fat in the diet, decreased adipocyte lipolysis and apoptosis, reduced body temperature and energy consumption) effect cicatricure the long-term influence on the body weight. Studies have shown that ghrelin levels correlate inversely with energy reserve, causing compensatory changes cicatricure in response to changes in weight. The growths levels due to loss pon
